What's an interesting route from London to South Wales avoiding the M4 and stopping off at interesting points?

You could take the M3, A303 to Stonehenge. Then follow the A303 to Glastonbury stopping off there. After that head to Bath before heading to Chepstow and the old Severn Bridge.

This will leave you in South Wales where you could follow the A48 to avoid the M4.

Others
While in London, get on a coach bus going west, and keep grabbing various buses and trains until you get there. You'll have an adveture, and really get to see a bit of those areas. Or you could grab your socks and sandals and just get a tour bus from London to South Wales.
